Manipulating “expression”, etc.

Detecting & measuring interactions

There are also many “biophysical” techniques I don’t have dedicated posts on, including:

  • surface plasmon resonance (SPR) (which detects interactions with molecules stuck on a chip)
  • isothermal calorimetry (ITC) (which measures tiny changes in temperature caused by binding interactions)
  • “single molecule” methods that use high resolution fluorescence microscopy or other techniques to track interactions

Experimental design & considerations

Another fundamental structural biology technique I don’t have a dedicated post on is NMR (nuclear magnetic resonance) – it’s basically a molecular MRI. It’s a solution phase experiment (so you don’t freeze your samples in place) that can work well for small floppy things, but requires lots of sample.
